Well the single version is how the song would have been released on Get Back , and the album version is the Specter mix that was on Let it Be.
The main audible difference is the different solos.

The main audible difference is the different solos.
Not so. The single was mixed by George Martin in 1970 and has a number of overdubs; the version on the ‘Get Back ‘ is far more the basic performance 31.64; Billy Preston’s swirly organ part is more evident in the mix for one.

I’ve often thought about that myself. I can’t sing worth a *** when I have a sore throat, let alone ten hours straight capped off by a real throat-shredder, so perhaps he didn’t have a sore throat specifically, just a head cold.

According to Lewisohn’s Complete Recording Sessions, that’s the way it was originally mixed and intended for the Get Back album. I don’t have the book in front of me (I’m at the office) but will check the page number and get back (no pun intended) to you.

Page 195: Sunday 4 January, 1970
It is widely believed that there are two different takes of ‘Let It Be ‘ publicly available – the single release (in the UK) on 6 March and the Let It Be LP version (ditto) 8 May 1970. Certainly the lead guitar solos in the middle eight differ considerably, and the LP version has a longer duration. But, in truth, these are one in the same version. That is, they derive from the same tape. By the conclusion of this 4 January 1970 session the eight track master tape had two lead guitar solos, playing side by side; the 30 April 1969 overdub and this new overdub [mentioned previously in the article].
The article goes on to say that in a remix for the single, the newer solo was mixed out and the first solo was mixed out for the album version. Unless further info is available that I am not aware of, the original intent (unreleased) on the master was to have both.
